Bacteriostatic water for Injection, USP (United States Pharmacopeia) is sterile water containing 0.9% benzyl alcohol as a preservative. It’s a clear, colourless, odourless liquid packaged in multi-dose vials. The benzyl alcohol is used specifically to inhibit bacterial growth after the vial is opened and punctured multiple times. It is used for research, as a diluent to reconstitute lyophilized peptides, proteins, and other research compounds. It is not for therapeutic use.

How does Bacteriostatic water work?

Bacteriostatic water works through the benzyl alcohol preservative.

  • BAC water can be injected into vial of powdered compound. It may dissolve the powder to create a liquid solution at your desired concentration.
  • Once you opened the sealed vial, bacteria may enter into it so 0.9% of benzyl alcohol may prevents those bacteria from multiplying inside the vial. It may stop replication, but does not sterile or kill the bacteria that were already present.
  • Due to bacteriostatic effect, one vial can be used for multiple withdrawals over the time up to 28 days after initial puncture if stored correctly at 2-8°C with proper sterile practise.

 Bacteriostatic water isn’t bactericidal, it does not kill bacteria it just prevents their growth and multiplication.  

Research Applications

  • Common research use is as a diluent for lyophilized peptides, proteins, and other compounds that require reconstitution.
  • Typical practice: Inject the desired volume of BAC water into the vial, gently swirl to dissolve. Use sterile technique for every withdrawal. Once opened, multi-dose vials are generally used within 28 days when stored properly at 2°C-8°C. For single-use applications, sterile water without preservative is preferred.

Research Dosage

  • There is no dosage of BAC water itself. The volume used depends entirely on the concentration you want for your research compound.
  • Formula: Desired concentration = Total mg of compound ÷ Total ml of BAC added
  • Example: 5 mg compound + 2 ml BAC = 2.5 mg/ml. Always calculate based on your specific research protocol.

Side effects

BAC water itself isn’t administered for effect. Side effects only occur if it’s used improperly or if benzyl alcohol is administered directly in high amount in that case side effects can be seen like burning, irritation, and pain etc. BAC water has no pharmacologic interaction as it isn’t active. Some compound is not stable in benzyl alcohol. Always check compatibility of your research peptide or compound with BAC water before mixing it. If you may combine multiple benzyl-alcohol preserved products then total BA exposure may increases.

Pharmacokinetics

Benzyl alcohol is metabolized in the liver to benzoic acid, then conjugated with glycine to form hippuric acid and excreted in urine. It has no therapeutic pharmacokinetic profile in this context because it is used only as a preservative/diluent at 0.9%. It may oxidize in liver to benzoic acid, then conjugated with glycine to form hippuric acid. Hippuric acid may get excreted in urine and the half-life of BA is approx. 1-2 hours in adult.
